Deletion of the mouse - globin regulatory element ( HS 26 ) has an unexpectedly mild phenotype

چکیده
Natural deletions of the region upstream of the human -globin gene cluster, together with expression studies in cell lines and transgenic mice, identified a single element (HS 40) as necessary and perhaps sufficient for high-level expression of the -globin genes. A similar element occupies the corresponding position upstream of the mouse (m) -globin genes (mHS 26) and was thought to have similar functional properties. We knocked out mHS 26 by homologous recombination and observed the surprising result that instead of the expected severe -thalassemia phenotype, the mice had a mild disease. Transcription levels of the mouse genes were reduced by about 50%, but homozygotes were healthy, with normal hemoglobin levels and only mild decreases in mean corpuscular volume and mean corpuscular hemoglobin. These results may indicate differences in the regulation of the -globin clusters in mice and humans or that additional cis-acting elements remain to be characterized in one or both clusters. Targeted inactivation of the major positive regulatory element (HS-40) of the human alpha-globin gene locus.
We have examined the role of the major positive upstream regulatory element of the human alpha-globin gene locus (HS-40) in its natural chromosomal context. Using homologous recombination, HS-40 was replaced by a neo marker gene in a mouse erythroleukemia hybrid cell line containing a single copy of human chromosome 16. In clones from which HS-40 had been deleted, human alpha-globin gene expres...
متن کاملDescription and targeted deletion of 5' hypersensitive site 5 and 6 of the mouse beta-globin locus control region.
The most upstream hypersensitive site (HS) of the beta-globin locus control region (LCR) in humans (5' HS 5) and chickens (5' HS 4) can act as an insulating element in some gain of function assays and may demarcate a beta-globin domain. We have mapped the most upstream HSs of the mouse beta-globin LCR and sequenced this region. We find that mice have a region homologous to human 5' HS 5 that is...
متن کاملRED CELLS Deletion of the core region of 5 HS2 of the mouse -globin locus control region reveals a distinct effect in comparison with human -globin transgenes
The -globin locus control region (LCR) is a large DNA element that is required for high-level expression of -like globin genes from the endogenous mouse locus or in transgenic mice carrying the human -globin locus. The LCR encompasses 6 DNaseI hypersensitive sites (HSs) that bind transcription factors. These HSs each contain a core of a few hundred base pairs (bp) that has most of the functiona...

The most upstream hypersensitive site (HS) of the b-globin locus control region (LCR) in humans (58 HS 5) and chickens (58 HS 4) can act as an insulating element in some gain of function assays and may demarcate a b-globin domain. We have mapped the most upstream HSs of the mouse b-globin LCR and sequenced this region. We find that mice have a region homologous to human 58 HS 5 that is associat...
متن کاملChromosome looping at the human alpha-globin locus is mediated via the major upstream regulatory element (HS -40).
Previous studies in the mouse have shown that high levels of alpha-globin gene expression in late erythropoiesis depend on long-range, physical interactions between remote upstream regulatory elements and the globin promoters. Using quantitative chromosome conformation capture (q3C), we have now analyzed all interactions between 4 such elements lying 10 to 50 kb upstream of the human alpha clus...
